Indiana University biologists have identified a hidden danger in the way bacteria build flagella, the rotating tails that enable movement. Their experiments used Bacillus subtilis, a harmless soil bacterium often studied as a model for disease-causing species. A bacterial cell wall normally prevents water from entering and bursting the cell. When the team removed PBP1, a protein thought to repair that wall, cells began dying precisely as their flagella formed. Further genetic tests showed that completing part of the flagellum activates enzymes that break down sections of the wall, possibly helping the structure fit and rotate. PBP1 usually repairs this damage quickly; without it, destruction outpaces repair and the cell splits open. The finding links movement machinery with cell-wall maintenance more closely than scientists had assumed. Because many harmful bacteria build similar structures through their walls, the vulnerability might eventually inspire drugs that disrupt infection, although the present study does not yet demonstrate a treatment.
