Oxford researchers have watched individual virus-like particles assemble molecule by molecule, overcoming a problem that previously left scientists with only snapshots or theoretical models. They studied an engineered particle made from 60 protein units. Mass photometry measured each molecule through the light it scattered, while a new confinement method kept the same particle in view long enough to be weighed repeatedly as it grew. The results explain how a structure can form reliably despite thousands of possible arrangements. Protein units first make weak, reversible contacts, so unproductive combinations can separate and try again. Successful closed shapes, however, contain several connections and become stable waypoints that guide later growth. A five-part ring marks an especially important early stage; once it forms, each subsequent stable stage requires fewer additional units, accelerating assembly. The technique could help researchers investigate antiviral strategies that interrupt capsid formation and support the engineering of vaccines or other therapies.
